- Aceasta biserica ramane mitropolia tarii din 1520 si pana in 1688 cand se muta la Bucuresti sub domnia lui Radu Leon. Despre minunata biserica, cronica tarii spune ca privind-o se satura ochii tuturor de vederea ei, iar Paul de Alep o descrie ca fiind cea mai mare biserica ce s-a construit pana la acea epoca si mai tarziu in Tara Romaneasca. Ca arhitectura se aseamana cu biserica mare a manastirii Snagov, zidita tot de Neagoe Basarab, lipsindu-i sinusurile laterale. Sub Constantin Brancoveanu (1688 - 1714) mitropolitul Teodosie a restaurat intreaga cladire, iar mitropolitul Antim Ivireanul reinfiinteaza in cladirile ei vechea tipografie in care se tiparesc carti de slujba si invatatura ortodoxa.
- Un secol si ceva mai tarziu, eteristii lui Alexandru Ipsilante i-au dezvelit acoperisul, topind plumbul pentru gloante. In periada urmatoare biserica este deteriorata considerabil. La 1842 s-au daramat toate cladirile care au format odata mitropolia Targovistei. Au ramas numai biserica si clopotnita de la intrare cu cele 4 clopote. In 1889 s-a hotarat daramarea ei iar intre anii 1892 - 1895 arhitectul francez Ize Compte du Nouy incepe constructia actualei biserici. Dupa daramarea vechii mitropolii, pietrele funerare de pe morminte au fost adunate intr-un muzeu al bisericii. Daramat si acesta in 1943, pietrele funerare au fost duse la muzeul istoric al orasului.
- Biserica actuala este construita in stil bizantin, in caramida, cu pridvor deschis, sustinut de 12 coloane. Este in forma de cruce greaca inscrisa. La intretaierea celor doua brate ale crucii se inalta turla naosului cu 12 laturi si 12 ferestre. Altarul are de asemenea o forma caracteristica, cu o absida mare in axul longitudinal si doua abside alaturate servind de proscomidie si diaconicon.
- Sculptura in lemn a catapetesmei este bogata si aurita. Pictura executata in 1927 de D. Belizarie in stil bizantin pe fond aurit ramane o lucrare de mare valoare a vremii noastre. Din fosta mitropolie se mai pastreaza o icoana de argint aurit cu capul Maicii Domnului si un epitaf din 1828 lucrat in plus rosu cu matase si fir. Astazi fosta manastire a Targovistei serveste ca biserica de enorie, fiind catedrala orasului.
